You pipette each of the components into a clean Eppendorf tube in the order below. You also remember to add a negative control without DNA. You also run a DNA sample from the victim to investigate whether the DNA from the T-shirt is the victim’s.

 

Water, nuclease-free 

30µL

EcoRI buffer

5 µL

DNA

10 µL (5 µg)

EcoRI enzyme

5 µL

Total volume

50 µL

 

You incubate the reaction at 37oC for 30 minutes. The reaction is stopped through thermal inactivation at 80oC for 5 minutes. 

You are anxious to find out the results and understand that you need to do a bit more work to visualize the results.
