You: The key is to find plates with a manageable number of colonies. Typically, we aim for plates with 30 to 300 colonies. This range provides a good balance between statistical accuracy and ease of counting. The colonies have to be well-separated to allow reliable counting, at the same time, if the sample is too dilute the results will not be reliable.

Sam: I see, in this case the 1/104 dilution seems to have fewer colonies than the required ones for statistical accuracy.
